High-Throughput PCR Product Purification Kit – 96-Well Format
Purification Technology
Purification is based on 96-well column chromatography, which allows DNA binding under high-salt conditions and elution under low-salt, slightly alkaline conditions. To begin, PCR products are mixed with a provided binding buffer at a 1:5 ratio. Mineral oil from PCR reactions does not need to be removed, as it is effectively separated during the purification process. The mixture is then applied to the 96-well plate using either centrifugation or a vacuum manifold. DNA binds to the plate based on ionic interactions, while primers, dimers, enzymes, and other contaminants flow through.
Bound DNA is washed twice with the supplied wash solution to remove residual impurities, and highly purified PCR products are finally eluted using the provided elution buffer. This process produces clean, high-quality DNA suitable for all common molecular biology applications, including cloning, sequencing, labeling, and enzymatic reactions.
